Engineering Clostridium Strain to Accept Unmethylated DNA

It is difficult to genetically manipulate the medically and biotechnologically important genus Clostridium due to the existence of the restriction and modification (RM) systems. We identified and engineered the RM system of a model clostridial species, C. acetobutylicum, with the aim to allow the host to accept the unmethylated DNA efficiently. A gene CAC1502 putatively encoding the type II res...

An atypical Clostridium strain related to the Clostridium botulinum group III strain isolated from a human blood culture.

A nontoxigenic strain isolated from a fatal human case of bacterial sepsis was identified as a Clostridium strain from Clostridium botulinum group III, based on the phenotypic characters and 16S rRNA gene sequence, and was found to be related to the mosaic C. botulinum D/C strain according to a multilocus sequence analysis of 5 housekeeping genes.

ClosTron-mediated engineering of Clostridium

Members of the genus Clostridium are of both medical and industrial importance. The molecular tools necessary to study and exploit their wide ranging physiological diversity through directed mutational analysis have until recently been lacking. The situation was transformed in the mid-2000s with the specific adaptation of intron re-targeting technology to the genus, through the development of t...

Differences in the accessibility of methylated and unmethylated DNA to DNase I.

DNase I binds in the minor groove of DNA and is used as an enzymatic tool to investigate the interaction of proteins with DNA. Here we show that the major groove located 5-methyldeoxycytidine can enhance or inhibit the cleavage rates of DNA by DNase I. This effect may be caused in part by changes in DNA structure affecting the accessibility of the minor groove of DNA to DNase I.

Engineering Hcp1 to Bind DNA

Current deoxyribonucleic acid (DNA) processing methods used for DNA sequencing and microarray analysis rely on restriction enzymes that produce oligonucleotides of heterogeneous lengths. Homogenous length DNA would be an advantage to chip based DNA analysis technologies as a way to reduce false positives (incorrect matches of complementary DNA). Nanotubes of discrete lengths could serve as a te...
